SBS2008 Domain.

I have successfully used the free tool and migrated 3 of our four workstation's user profile.

The last one is fails. The progress log shows:
Binding to Active Directory ...Fails.
There are no more endpoints available from endpoint mapper
Migration Fails.

During migration attempt (Binding to Active Directory...)a dialog box open for credentials for a domain user name and password. I entered in the domain admin credentials and still it fails.

Verified the computer is on the domain, I can browse shares etc

This isn't a User a User Profile Wizard problem. The most likely cause is a DNS or network issue; you should certainly check the IP configuration of the machine and check whether it is different to the machines you have migrated successfully.
 
There is a Microsoft article here. This relates to Server 2003 buit is still relevent.

can I use Profwiz Wizard in Windows 2000

You need to use the Windows 2000 Compatible version available on the Support Downloads page.
